Enterprise System EssayThis report analyses a fast moving goods guilds information systems. The report analyses the conjunctions major types of information systems, including a more than detailed assessment of its Decision Support System and issues relating to hardware and software technology and telecommunications. The political party that will be analysed in this report is a supplementary of large international fast moving consumer goods company. The business of the subsidiary company is the bottling and selling of mineral water in Vietnam.Information for this report is based on 2 interviews held with the recently retired Chair of the company (to be referred to as the Chair). The Chair had some reservations about being interviewed for this assignment due to possible commercial in confidence issues. Ron Williams agreed that neither the actual names of the subsidiary company (to be referred to as FMCG Ltd nor the international fast moving goods company (to be referred to as world(prenominal) Ltd) have to be used in the assignment. The report also includes an analysis of the Canberra Institute of Technologys website.Question 1 (a) Introduce the organisation. Describe the organisations features much(prenominal) as business processes, culture, structure, environment, business strategy FMCG Ltd has been operating for some 6 years in Vietnam and was the first entrant into the mineral market there. FMCG Ltd is managed separately from International Ltd and has an entrepreneurial culture and a flat management structure. It is highly profitable. The spring water market in Vietnam is now saturated, with a number of imitation products accessible locally. This makes further growth in market share by the company difficult to achieve. The companys main strategy is to append profits through efficiencies in its bring home the bacon chain. This has been achieved, in part, due to an improvement in business Information Systems (IS) through the establishment of an En terprise System, which will be discussed in this report. The information system enables the company to maintain close control of the supply chain. FMCG Ltd has recently introduced its own sales staff to supply the retailers of its products. This has been critical in ensuring the products are effectively placed and promoted. This has enabled the company to increase its sales dramatically. The use of the companys own sales force allows important sales information to be gathered and fed into the companys information system. This information assists the company, the wholesalers and the retailers. The company closely supervises and assists the wholesalers, retailers and the company that is responsible for collecting the mineral water at its source. The Chair said that the Enterprise System is the heart of the company. The company uses the system to increase the companys efficiency and profitability. Fahy discusses the benefits of strategic enterprise systems (Fahy, 2001). He emphasises the importance of the management issues as well as the technical challenges associated with introducing such a system. The Chair said that he agreed with this comment. The Chair also said that the benefits of such a system as outlined by Laudon (Laudon, 2002) were experienced by FMCG Ltd .

My Critical Views On Global Climate Change Essay Example for Free

My Critical Views On Global humour Change EssayIt is receive that even contempt actions with regards to using measures and activities that are environmentally friendly, much more work is needed in order to address the problem of global humor change. The implementation of supranational protocols and requiring countries to meet certain environmental standards is certainly a big step towards addressing this issue. More intergovernmental cooperation is also a nonher prospective answer to this problem. The more developed countries should also take the lead in encouraging the shift away from more traditional methods of production towards more environmentally friendly methods through the award of grants and change over of technology (Torn and Harte, 2006). The creation of a body that is designed to not only specifically develop policies regarding this issue but also be given the forefinger to enforce these policies will allow for the more efficient reduction of these nursery gas es (Torn and Harte, 2006).It is unclear just how effective these policies may be but the fact that is clear is that in order to address this issue, each and every person in the world must take part in reducing the emission of these greenhouse gases which is the main cause of this problem (Gore, 2006). Simple steps such as taking the public transport or walking instead of taking coke monoxide emitting vehicles will go a long way. there are many things that people can do in order to contribute to the improvement of the certain environmental condition. It does not take a lot and it all starts with the awareness of the gravity of the situation. While the general scientific consensus is that global warming is real and its boilers suit effects are detrimental, there are distillery some prominent scientists who feel that the severe effects of global warming have been sensationalized. Certain members of high society take the idea of global warming with the normal temperature and that the temperature of the surface of the Earth has not severely increased over the last one hundred years. They pack that the temperature data collected in the 1970s were taken in urbanized areas using different kinds of thermometers, hence the temperature readings were influenced by human error and do not precisely show drastic temperature changes in the surface of the Earth as well as in the oceans. It is therefore important the satellite data be acquired for the coming next decades in order to generate robust and reliable temperature readings that society and the scientific world will acceptable without any doubts on its believability and efficiency. It is also of prime importance that scientific and legislative bodies prove that global warming does actually occur as a result of carbon dioxide gathering in the atmosphere. Geological scientists have gather substantial fossil evidence that prove that carbon dioxide concentrations have increased on the surface of the Earth, which thus results in a warmer surface, but the direct connection between the increase in carbon dioxide and the temperature increase at the actual in situ settings should still be observed. Most of the information and generalizations that scientists have collected were based on laboratory conditions where most of the components of the setup were easily regulated and controlled, such as humidity and temperature. There are currently a number of models that may be used in forecasting average surface temperatures on the Earth, but there is still a need to also design a model that could predict the amount of human-made emissions that could trigger the same features that cause global warming. There is quite a huge number of people in society that still do not understand the climate changes and what triggers such disturbances in our current weather. In the join States, there has been some restriction with regards to releasing information and disseminating information with regards to global warming. T he Union of Concerned Scientists to the Oversight and Government Reform Committee of the senate has reported that the current administration has instructed research scientists to filter any information that they release to the public and that only issues that fit the current administration should be reported. The phrases of global warming and climate change have thus been identified as taboo in any type of communication that is delivered to the American society. Such restriction has resulted in the calmness of a number of scientists who have a personal belief that the truth with regards to any discovery should be released to the public as soon as realistic (Andrew, 2007). Every country is now aware of the causes and effects of global warming on the ecosystem. There has been great speculation that global warming is both a natural and a man-made phenomenon.Whatever the cause is, it is imperative that every citizen understands what global warming is and what measures can be taken in o rder to minimize, invalidate or prevent the increase in accumulation of greenhouse gases in the atmosphere.References Andrew B (2007) Scientific bodies must take own action on emissions. personality 448(7155)749 Choi O and Fisher A (2003) The impacts of socioeconomic development and climate change on severe weather catastrophe losses Mid-Atlantic region (MAR) and the U. Simone De Beauvoir: Gender and Sex

Simone De Beauvoir Gender and SexSimone de Beauvoirs Insights Towards Gender and StatusIs in that location any wonder that Simone de Beauvoir rejected the idea that genderhas im gayent char croperistics from birth? As a womanhood philosopher living in a mans gentlemans gentleman I believe she fully understood what she was public lecture intimately. The writings in her world ren have goted book The Second Sex written in 1947 give us a historical and cultural lesson of what women give birth had to overcome to exist as women in a mans world. work force and women be both influenced by their conditioning the main difference is that man has been attri neverthelessed the head teachering role. Times ar changing and Simone de Beauvoir has played an important role in our attempt to undo the erroneous programming. Todays leading ladies choose proven their capacities at playing the part.Simone de Beauvoirs example of what a woman that is intelligent can do in demeanor by her straight forward, honest woo may be the stepping stones for other women to disquietudelessly do the said(prenominal). As far back as the 10th century ideas of antifeminism mystify haunted the women of the world. The handful of women that were in the limelight were usu all(prenominal)y obliterated for witchcraft as in the case of Jeanne D run or publicly ridiculed or at worsened martyred. Women as the lesser sex have been depicted as temptresses from Bible days which at the same cartridge clip undermined their intelligence. This dual image which has conditioned women to perk up themselves as passive beings who lie in wait for men to save, them or come and love them and their counterparts the devouring temptresses who defend what they want and leave the rest arsehole including their intelligence is what Simone de Beauvoir sees as our conditioning. This conditioning starts at a in truth early age when there argon no app atomic number 18nt differences between go bad girls and baby boys. They argon both at this early age totally in need of nurturing and forcible contact which they thrive upon. This loving care is exactly the same up until a certain bite when the baby is weaned. According to Simone De Beauvoir from that point on boys are taught to sire little men and they receive less physical contact and affection than their girl counterparts They are also a product of their conditioning just as we are. This is an important thing to remember throughout this reading. Boys and girls resembling are part of their environmental conditioning through family heritage, cultural traditions, religious beliefs and historical beliefs. As children we all have the same attitudes of temper tantrums, pouting, charming our parents thence we are in fact the same until our environment gives us a nonher outlook upon ourselves. Little girls quickly learn that they need to be careful. They need to act like little ladies non climbing trees and skinning knees as their brothers are enco uraged to do. This myth starts very early with the family attitude about the physical differences between sexes. Boys have an outward expression of their masculinity which turns them rapidly into having something to be proud of. This expression of self that has, in the eyes of the parents, a value compels what the boy measures himself to other boys with. Its a tactile indicator of their manliness and gives them the instinct of measurability. How long their sex is and how far they can urinate. The inevit adequate presence of their extension gives them a power of object. Women are inwardness. All of their discoveries are inward and therefore left unsaid. You cant see much of the sex of a little girl so it is non do a fuss over as boys sexes are. Girls try to express their transfer towards an object to a gentlewoman for example. This treating a doll as herself gives her the idea that the substance is the same and therefore she is someone to dress up to cater to, that has an humank ind because others make her exist. Later on in life she will eventually become that doll thing that sits home waiting for her husband and children to pick her up and do as they please with her. Just as she idled away her childhood she idles away the hours of her parenthood, or wifehood at home staving off boredom with no particular goals or worlds to conquer.Boys grow up with the idea that they are going somewhere and have something to accomplish and will assume responsibilities along the way. They learn at around 13 yrs. old that the answer may not come out in words but through violence. A girl wills repress her own violence because girls dont do that cast of thing. This violence turns into neurosis which in part is violence unexpressed which comes back to oneself eventually. Simone De Beauvoir was avant guard when she recognized the fact that women have their own part of violence at that age but we do not allow ourselves to express it. We are simply not encouraged socially, histo rically or culturally to express it. Where age has been made is that more and more women are training in martial arts which are a positive way to express their inner violence. What brainsick our lets, and depending on what conditioning you were also given, was the pre- established path chosen for us no matter what we would like to become. Once the path was set then fear was quickly to follow. What path? The path that says no matter what education you receive you will end up a wife and mother then grandmother. This is a frightening aspect of the growing girls plight. She will not only have to learn to become a woman, a wife, a mother and a grandmother. She can study all that she would like but there is no real asking of her to do so or to assume the responsibilities that this implies. Shell attach then stop working to have children and her husbands career will prevail and take on the traditional role of breadwinner. Perhaps straightaways economy has obliged young women to be care er assertive but maybe not for the right reasons. Career minded because families are not with expecting without two good salaries to support them. The outer expression of sexual pride may still be baring the road. The fact that a young woman can be a productive and important member of our society and that she also has her contribution to make does not seem to impress upon girls as it does on boys. There are so many frightening biological events in the life of a young girl who is becoming a woman that possessing her own inward sexuality is a complicated task.Young men can see theirs, be proud of it, and use it where as young girls have to plenitude with what biologically is imposed upon them. Their menstrual cycles which alter their characters for several days a month is in itself part of the mystery of womanhood. The deviates in her body that may lead her to withdrawal because now she is becoming the object so desired by men. The doll has come of age and can now be manoeuvred accor dingly. This is a moment in time when many young women become anorexic keeping their bodies at bay and stopping the menstrual cycle as if time would stand still. The onset of an eventual pregnancy, abortion risk, risks during pregnancy, menopause and all the plectrums that implies has made being a woman probably more complicated than being a man. What is very interesting to remark according to statistics there are a large absolute majority of women that would prefer being men. Men in a very exquisite minority if they had the choice would have preferred being a woman. There is the unfortunate, radical, feminist view of continuously proving that women can be more or break off than men by doing the same things and acting the same way.. I find this unfortunate. If we want friction matchity then we should be striving for equal respect and equal rights not superiority and domination which are what has made us suffer from the ages. Women finding ways to be assertive, well-balanced, br illiant, and effective is what can give us our qualification in todays world. It was unthought-of at the time of French sculpt rice Camille Claudel that a woman could surpass the master. Camille Claudel would never have low-pitched through the layers of men to get to where she did in the arts if she hadnt had the support of her father firstly and Rodin. Once Rodin retracted she lost all prestige and crack over her life. There is the great Mexican artist Frida Kahlo who also had the support of her father interestingly enough and that of a great Mexican artist as well. Both fathers in these two examples gave their support to their daughters which were probably what they would have received if they were sons instead of daughters. In the face of talent and genius these men were supportive and able to recognize potential. How many other Sunday morning painters got left by the roadside because they were not recognized in virtue of their talents but because of their gender? What did the w orld wars teach us about women? It taught us that in the absence of brothers, cousins, fathers, and husbands they were capable of running a country from industry to education the national health Women kept their families fed, clothed, and sheltered during the war. We are not warriors are heart but have the same potential. There are examples in hi narration like Amazons and Jeanne dArc of women capable of leading armies and warring. We could have been cultivated as war lords also but werent. Women are givers of life and that is probably what gives us the incentive to promote quiet culture instead of war. No woman wants to see her sons, daughters, husband, brothers or sisters go off to war and not come back. The peace movement has a majority of women supporters and that seems to go with the fact that life giving is still our realm. The political world has opened up its doors to women within the last twenty years. Before that time there was usually a woman behind a great man in the sh adows and there to decorate the photographs for the press more than for her opinions. In France there were three woman ministers elected in the late 70s. This was a turning point. The paradox comes in when women fought in the French resistance showing their intrepidity and didnt obtain the right to vote until two years after the war was over. The equality act was also pronounced shortly after womens right to vote. This maintaining of women in a conditioned status has been painful for us and is now becoming painful to our male counterparts who are seeking the limits of their own identity. Women are now brought up and beginning to think for themselves, what they want out of life, what they are going to do to get what they need. This is a true cultural revolution. If the conditioning has begun to change then women shall become women and know themselves. Thus knowing themselves they will become decision makers, and active citizens not through mans making but through their own. This fun damental step has been operating since May 1968 even though precursors since ancient times have been trying to change this. Simone de Beauvoir succeeded in starting the partnership for Womens Rights and was a militant for family planning.Giving back to women the right to do what they need to do with their own bodies was an essential step in accepting the responsibilities that are incumbent for every human being either man or woman. The big waking up of our society started with her book and has continued to live on inspiring women to take the responsibility for their lives as a person and an equal which gives them a lot more freedom than it did our mothers. The social role was encouraging us look pretty but please dont say anything. Mediocrity amongst woman rose as her only idea of self was stemming from what others mirror her. Her husband and her family were her only mirrors and connection with the world. He went out in the world she quietly stayed home or followed in the shadows. If an intelligent woman did express herself she was ridiculed in public and could not be accepted for her thoughts. The Marie Curies and other thinkers are a small handful of the potential that women have been suppressing since the beginning of time. Women are beginning to realize that no matter what their conditioning since birth they can change the course of events in their lives. Taking their own responsibility for their own lives and what is to become of them is the next chapter in our evolution. Women are finally starting to be raised to believe in themselves and their capacities. Ask a little girl today what she wants to become later and she might surprise you and answer an astronaut, a research worker, an architect . We are far from the days of Simone de Beauvoirs struggle to live in a mans world. I think that men are having trouble adjusting to this change. They are becoming more and more aware of womens presence in the work field and their contribution as fellow citizens. T heir own identity has been questioned and ours is beginning to grow and develop. This cleavage will be difficult to stabilizesince all related gender problems have finally come to the surface, the debate is whether or not women that are raising their children can make the shift to raising their sons and daughters alike. The story perhaps begins and ends with women. Are we going to keep attributing to ourselves the role models that went before us? Or are we finally going to break the bondage of what we have been made to think of ourselves? There has come a time when the individual man or woman will begin to raise his consciousness and choose him or herself first. Not in and egotistical way but to ask oneself the question What do I need for myself right now to be in musical arrangement with myself? When finally women and men have worked on themselves enough to be able at any moment to ask themselves that question then it is definitely not a question of gender but autonomy. Do women w ant their autonomy? Is the calorie-free way out or women to continue a mediocre existence to take the easy way out and let this be a mans world and a state of dependency and submission? Men take their independence and unfortunately women think or have been taught to think theirs is to be earned. Men believe it is their right from the beginning. Women ask permission for it and in just about circumstances and it never comes. Leaving behind all the social prejudice and family prejudice and historical and cultural prejudice can only be the choice of women. Simone de Beauvoir showed us that a woman is herself and can dare to be herself if she so chooses. Although this idea will not appeal to some, women are running for presidential maculation these days and hold important functions within our highest auspices while Simones message has probably hit home. Women are accepting to be themselves without asking permission to do so. Men are battling with their own conditioning and their own identity problems due to this conditioning. In order to create a balance it will take time and courage. Women for peace, for childrens rights, for non-violence, for equal education and career rights have united all over the world to speak their piece. A beautiful French numbers that implies women as being mens future. could hold a lot of truth in these words.Thinking back on the historical events and cultural ideas about gender it must have taken great incentive for a woman like Simone de Beauvoir de openly and honestly explore the femaleness and the contradictions of our times. It is also easy to comprehend why men find it so difficult to understand us when we are on the brink of finding ourselves as people and as women. Perhaps if our own opinion of self changes then the worlds opinion can also evolve. Starting with individual awareness can only lead to popular awareness in time. Simone de Beauvoir in her rejection of gender has put back the sparkle in the eyes of women and help ed us overcome the conditioning of the years.ReferencesSimone de Beauvoir Le Deuxieme Sexe Galiard, Paris 1947Nancy Bauer subscriber Simone de Beauvoir Philosophy and Feminism Columbia University press N.Y. 2001Isabelle De Courtivron Contributor New French Feminism An Anthology ED. Effect of Curcumin on Periodontis

Effect of Curcumin on PeriodontisINTRODUCTIONPeriodontitis is a chronic incendiary disease characterized by wipeout of the soft and hard create from raw stuffs encouraging the tooth. This tissue destruction appears to outcome from complex interaction between the bacteria and the droves immune and inflammatory system. The horde immune repartee identifies these invading pathogens and tries to neutralize or kill these microorganisms. During this process the host response elaborates a variety of mediators such as pro-inflammatory mediators, cytokines and proteolytic enzymes such as matrix metalloproteinases (MMPs). It is now believed that the major component of the soft and hard tissue destruction seen in periodontal disease occurs as a resolving of activation of these mediators.1 Thus this host response which is essentially protective by intent paradoxically can too result in tissue disablement. accordingly late(a) therapeutic efforts are now focusing on altering or modula ting this host response. This lead to emergence of a new comprehensive sermon strategy for periodontitis which involves Host modulatory therapy (HMT) which can be combined with traditional periodontal therapies.2To date, there is one FDA approved, general therapy that is prescribed as a host response modifier in the treatment of periodontal disease and that is adjunctive subantimicrobial dose doxycycline (SDD), which down-regulates the practise of MMPs.3These MMPs are considered to be primary proteinases involved in periodontal tissue destruction synthesized by a variety of infiltrating inflammatory cells (i.e. neutrophils and macrophages), resident cells and some bacteria. Numerous studies deem substantiated the relationship between MMPs and periodontal diseases, with findings of strongly higher MMP-9 (neutrophil derived MMPs) levels in patients with periodontitis compared to healthy controls.4, 5 Doxycycline is the most potent tetracycline for ban of these MMPs.6 However d our term tetracycline therapy has veritable disadvantages, the most important being the development of microbial resistance to this medicate.7At preface a health care system exists in India where different theatrical roles of diseases are being treated by Ayurvedic method. Since ancient times Ayurvedic drugs are being utilise for the treatment of various diseases, including periodontal and other oral diseases.8 They are widely acclaimed for their minimal side- motions and cost-effectiveness. Use of herbal tea prolongs in treating periodontal diseases does not produce side cause of tetracycline compounds as well as other unreal drugs.Curcuma longa (Turmeric) is a widely used herb which is distributed throughout tropical and subtropical regions of the world and widely cultivated in India, China and Indonesia.9 It has also long been used in both Ayurvedic medicine and Chinese medicine as an anti-inflammatory drug agent.9 The sprightly constituent in turmeric is Curcumin.9, 10 Curcumin is known to rent an anti-inflammatory effect. Studies show that Curcumin limits the activity of two enzymes, lipoxygenase and cyclooxygenase-2 that are involved in promoting and maintaining inflammation.11, 12 Studies strike also shown that Curcumin down-regulates the MMP-9 activity.13, 14 Thus it may accept a potential role in the treatment of inflammatory periodontal disease. Its anti-inflammatory activity in periodontal diseases has not been extensively investigated. With this downplay, the meditate was conducted to value the anti-inflammatory effect of Curcumin by its inhibitory effect on MMP-9 activity and compare the same with Doxycycline, which is known to have an anticollagenase activity.MATERIALS AND METHODSSubjectsA total of 30 sites from 30 subjects (males and females), above 35 yrs of age, diagnosed clinically as having Chronic Periodontitis with pocket depths ranging from 5-8mm were selected for the study. The patients had no history of pervious dental tre atment, antibiotic or anti-inflammatory drug therapy for the past 6 months. Patients with any history of systemic diseases / conditions, pregnant patients and smokers were excluded from the study. An informed consent was obtained from the patients prior to their enrollment into the study, and the study has been independently reviewed and approved by the institutional ethical board. Post collection of samples, the patients who participated in this study received the standard treatment protocol for Chronic Periodontitis.Method of Collection of SamplesGingival tissue samples were obtained infra aseptic conditions after administration of local anesthesia from patients who were diagnosed with Chronic Periodontitis. Gingival tissue samples were obtained from sites with pocket depths ranging from 5-8mm. Immediately following excision, these tissue specimens were briefly washed down the stairs cold distilled water and blotted dry. Then the specimens were transferred into sterile plastic v ials which contained Phosphate Buffer Solution (PBS), pH 7.2, that acted as the transport medium. Specimens were then immediately carried to the laboratory where they were stored at -80C until use.IN-VITRO STUDYChemicals UsedPure extract of Curcumin in powder form (Samy Labs, Bangalore, India) and Doxycycline i.e. pure doxycycline hyclate in powder form (Aristo Pharmaceuticals Limited, Bangalore, India) was used for the purpose of the study. The Curcumin stem was prepared by dissolving 15mg of Curcumin in 10ml of distilled water (1500 g/ml). Preparation of Doxycycline solution was by dissolving 3mg of Doxycycline in 10ml of distilled water (300 g/ml).Extraction of MMP-9The frozen gingival tissue samples were allowed to thaw to room temperature. Then each(prenominal) tissue sample was homogenized with 2.5% Triton X-100. This was followed by centrifugation of the homogenized samples for 30 mins at 6000 rpm, at 4C. The resulting supernatant was separated and used for analysis.Detecti ng the Optimal Curcumin and Doxycycline ConcentrationTo determine the optimal inhibition of MMP-9 by Curcumin, different preoccupations of Curcumin i.e. 300, 500, 1000, 1500 g/ml were added to 50l of gingival tissue extract and incubated at room temperature for 60mins.The increment of increasing concentrations of Curcumin to the gingival tissue extract resulted in decreased MMP-9 activity as shown in Figure 1.Assessing the MMP-9 activity by Densitometric Analysis demonstrated that the concentration of Curcumin required to inhibit more than 50% of MMP-9 activity was 1500 g/ml. Since Curcumin showed more than 50% inhibition of MMP-9 activity at a concentration of 1500 g/ml as compared to the control (without drug), this concentration was used to pretreat the gingival tissue extract in the study. The Curcumin solution with a concentration of 1500 g/ml was freshly prepared prior to processing of each sample of gingival tissue extract.Similarly increasing concentrations of Doxycycline ie 50, 100, 200, 300g/ml were added to 50l of gingival tissue extract and incubated at room temperature for 60mins. Assessing the MMP-9 activity by Densitometric Analysis demonstrated that the concentration of Doxycycline required to inhibit more than 50% of MMP-9 activity was 300g/ml and thus this concentration was used to pretreat the gingival tissue extract in the study. The Doxycycline solution with a concentration of 300g/ml was freshly prepared prior to processing of each sample of gingival tissue extract.Pretreatment of Gingival Tissue Extract with Curcumin and DoxycyclineTo compare the inhibition of MMP-9 activity, 50l of gingival tissue extract was preincubated with freshly prepared solution of 50l of Curcumin (1500l/ml) and 50l of Doxycycline (300l/ml) solution for 60mins at room temperature in separate vials. In addition, 50l of extract was also incubated with 50l of distilled water which was used as the control. verification for MMP-9 ActivityThe presence of MMP-9 activ ity in collected samples was studied by gelatin zymography. Sodium Dodecyl Sulphate-Poly Acrylamide Gel Electrophoresis (SDS-PAGE) on 10% polyacrylamide containing 10% SDS copolymerized with 1g/l gelatin was used to pretreated tissue extracts under non reducing conditions without prior boiling. After electrophoresis, in order to regain the enzyme activity, the gels were rinsed with 2.5% Triton X-100 for 1hr to remove SDS thus allowing the protein to denature. The gels were then immersed in a proteolysis buffer containing Tris- HCl 50 m M/l (pH 7.6) and CaCl2 20mM/l and incubated at 37 C for 16 hours. The gels were subsequently stained Coomassie Blue (0.25% Coomassie undimmed Blue R250, 40% methanol, 10% acetic acid). Gels were destained (30% methanol, 10% acetic acid and 60% water) until white bands appeared clearly from the blue background. These bands of gelatin lysis detected against the blue background as seen in Figure 2, represented enzymatic activities.RESULTSThe presence of MMP-9 was studied. The enzymatic activities were detected as unstained bands on gelatin gel by zymography technique. In order to measure the relative MMP-9 levels, multi-image gel documentation systems were used to scan the clear zones and the fortune of inhibition was analyzed. Significant differences were found in the MMP activity in treated groups compared to the control.Figure 3 illustrates the step-down in the MMP-9 activity of each sample of gingival tissue extract when incubated with Doxycycline and Curcumin, expressed as percentage of reduction from the control.Table 1 shows the range, mean values and the standard deviation (S.D.) values for Curcumin and Doxycycline. The mean values show 59.58 % reduction in the MMP-9 activity with the addition of Doxycycline and 61.01% reduction in the MMP-9 activity with addition of Curcumin to the gingival tissue extract under identical conditions.Table 2 shows comparison of MMP-9 inhibitory effect between Doxycycline and Curcumin. The results obtained were analysed statistically using students paired t- test.DISCUSSIONPeriodontal diseases are the result of destruction of the soft and hard connective tissues surrounding and supporting the teeth. Inflammation is the basic cause of initiation of this breakdown. Research in the field of pathogenesis of periodontal disease has shown that various enzymatic activities which are directed towards the destruction of the pathogen also result in destruction of the periodontal connective tissue. Research has shown that, Matrix metelloproteinases (MMPs) are one group of enzymes which are responsible for this destruction to a astronomical extent. MMPs are involved in a number of physiological events including, tissue remodeling and pathological diseases such as periodontal disease and are the major players in collagen breakdown during periodontal destruction. MMPs are normally tightly regulated and disruption of this regulation leads to the pathologic breakdown of connective t issues. Higher levels of MMPs in the periodontal tissues annoy an imbalance between the production and degradation of collagen, causing tooth attachment loss. Especially, PMN derived MMPs (MMP-8 MMP-9) are the main proteinases related to tissue destruction and remodeling events in periodontal diseases. Numerous studies have substantiated the relationship between MMPs and periodontal diseases, with findings of significantly higher MMP-9 levels in patients with periodontitis as compared to healthy controls.4, 5, 15With this understanding of the role of host immune response in periodontal destruction the possibility of host modulation so as to reduce the destructive aspects of the host response and hence reduce the damage to the periodontium was investigated. First group of drugs that showed this host modulation activity were tetracyclines. Studies have demonstrated that tetracycline could significantly inhibit collagenase activity in GCF and gingival tissue, even at lower social di sease than traditional antimicrobial dosages ie sub antimicrobial dosage.3 Tetracyclines inhibit collagenases by binding to the Ca2+ or Zn2+ (cations) required for the activation of MMPs such as collagenases and gelatinases.16 It is also possible that tetracyclines can inhibit synthesis of neutrophil-derived oxygen radicals, suppressing neutrophil migration and degranulation.17 However, microbial resistance to tetracyclines has been reported.7 In addition, long term treatment can result in other side- set up such as anorexia, nausea, epigastric distress and fatty liver. A few recent studies on collagenase inhibition suggested that, like tetracyclines, herbal extracts are also potent inhibitors of pathologically elevated collagenases and hence they may be used as an alternative adjunct in the management of periodontal diseases.18, 19 Ayurvedic drugs like Neem, Triphala, Bakul etc have been used therapeutically since ancient times to treat diseases, including periodontal diseases. Wi th the advent of modern synthetic drugs their convenience of standardized dosage form, dramatic efficacy in acute conditions and most of all simplicity of usage, there was a decline in the use of the plant medicines. But a long term treatment, with these synthetic drugs, has many adverse effects and they are also not cost effective. As Ayurvedic drugs, are widely acclaimed for their minimal side-effects and cost-effectiveness in India, they are now again being used extensively in treatment. Use of herbal extracts in treating periodontal diseases does not produce side effects of tetracycline compounds as well as other synthetic drugs. It has been shown that certain ayurvedic medicines also have the host modulation effect, similar to that of tetracyclines. A few recent studies conducted on collagenase inhibition by herbal extracts suggest that, like tetracyclines, herbal extracts are also potent inhibitors of pathogenically elevated collagenase and hence may be used as an alternative a djunct in the management of periodontal diseases.18, 19, 20Turmeric which has Curcumin as its active constituent has been shown to have anti-inflammatory, anti-oxidant, anti- bacterial, anti- viral activities etc.10 Its anti- inflammatory activity has not been investigated in periodontal diseases. With the above details in mind the present study was conducted to evaluate the anti-inflammatory property of Turmeric. In the present study the anti-collagenase activity of Curcumin was compared with that of Doxycycline which has proven anti-collagenase activity. In the present study we chose Doxycycline because it has been proved that it is a more potent collagenase inhibitor as compared to minocycline and tetracycline.6 Studies have shown that Curcumin down-regulates MMP-9 activity13, 14, 21 and thus, in the present study we chose Curcumin to find out its anti-collagenase activity by checking its effect on MMP-9 activity.Research has shown that the predominant MMPs in inflamed gingival and periodontal tissues are PMN- type MMPs (MMP-8 MMP-9). Elevated activity of gelatinases (MMP-2 and MMP-9) has also been found in inflamed gingival tissues from chronic periodontitis patients.5, 22 The activity of PMN- type MMPs has been found not only to positively correlate to the severity of inflammation and pocket depth but also the amount of these enzymes recovered in an active rather than latent form appears to increase with greater severity of periodontal disease in the gingival tissue.20 Thus, since the above studies have shown that activity of PMN- type MMPs is increase in inflamed gingival tissues, in the present study we decided to use gingival tissue samples that were obtained from the patients diagnosed clinically with chronic periodontitis. The reduction in the MMP-9 activity of each sample of gingival tissue extract when incubated with the drugs .i.e. Doxycycline and Curcumin, was expressed as percentage of reduction from the control (without drug). The mean values sho wed 59.58% reduction in the MMP-9 activity with the addition of Doxycycline and 61.01 % reduction in the MMP-9 activity with addition of Curcumin to the gingival tissue extract under identical conditions. These results show that Curcumin has a significant inhibitory effect on PMN-type MMP-9, and this inhibitory effect of Curcumin is comparable to that of Doxycycline when the percentage of inhibition as compared to control was analysed. Curcumin also showed a significant reduction in the MMP-9 activity with an average inhibition of about 61% which is also statistically highly significant (p 23 and 12 g per day.24 But the boilersuit results show that Curcumin has the ability to significantly inhibit the MMP-9 activity. The present study suggested that Curcumin could produce significant inhibition of MMPs at 1500 g/ml concentration, which is well under the gumshoe drug profile confirmed by toxicological studies.23, 24 Thus this study shows that Doxycycline and Curcumin possess anticoll agenase activity in vitro. The finding of the present study is significant and confirms the use of Curcumin in treating periodontal diseases. A recent in vivo study showed that Curcumin modulates periodontal disease and had potent anti-inflammatory effects when it was systemically-administered in ligature-induced periodontitis in rats.25 Studies have shown that one of main mechanisms for the anti-inflammatory effects of Curcumin may be the inhibition of NF-B (nuclear factor kappa B).13, 25 The Curcumin-mediated inhibition of MMP-9 gene rumination appears to occur via NF-B and AP-1 (Activator Protein-1) because their DNA binding activities were suppressed by Curcumin.14, 21When compared to tetracycline, Curcumin has better anti-inflammatory effect, is more cost effective and has minimal side effects and thus can be tried as a substitute for Tetracycline as an anticollagenase agent. Curcumin also allows suppression of collagenase activity well within the adept dosage profile confirme d by toxicological studies. The in vivo environment is substantially different, and inherent limitations of an in vitro replication may constrain our understanding of the systemic effects of Curcumin. Further in vivo studies of Curcumin are needed to define its toxicological profile before making it a part of the therapeutic regimen in periodontal treatment.CONCLUSIONIn the light of observations from the current study it can be concluded that Doxycycline and Curcumin possess anticollagenase activity in vitro. Since Curcumin has better anti-inflammatory effect compared to Tetracycline, Curcumin being more cost effective and with no side effects can be tried as a substitute for Tetracycline as an anticollagenase agent. Thus Curcumin may have therapeutic potential as a host modulation agent in periodontal diseases.

Blockbuster Video Analysis Essay -- essays research papers

The television system rakehellal industry was one of the more growing serve retailers in the mid-1990s. However, due to Blockbuster, many rental video companies have failed to compete against this category killer. wolfram Coast Video, Video City, and Hollywood Video, which are among the few and just now large competitors of Blockbuster in the tri-state area. Many family-owned video rental stores could not compete against Blockbusters assortment of videos. Blockbuster opened its first store in 1985 and has grown to become the worlds number one video chain. Blockbuster is open 365 days a year with 9,800 stores worldwide. Customer base has risen above 50 million in the United States as well as several million more worldwide. The Gallup Organization, known for the Gallup Polls, declared that Blockbuster has 100 percent recognition. Most importantly, 70 percent of the population in the United States lives deep down cardinal minutes of a Blockbuster store. Category killers, howev er, usually rend customers from far distances. Because most customers are ten minutes away, small businesses in competition will fail. A modern marketing campaign has been the promotion of DirecTV, a digital satellite service election to cable television. Also, an alliance with Radio Shack has been in effect promoting new technologies. To compete against the up rise of online rental services, Blockbuster has been testing online rental services which allow customers to reserve and rent videos from specific stores. The other services retailer is West Coast Video, a Philadelphia-based company that was once a supreme competitor to Blockbuster. West Coast Video was at one point, New Jerseys largest video rental chain in 1997. That preceding year, the company acquired stores from Palmer Video, tops(p) Video, and Great American Video. It became the fourth largest video chain with close to half of its 568 stores in New Jersey. However, the company has approximately 225 stores to avoid going out of business.
